Addition of residues and reintroduction of microorganisms in Jatropha curcas cultivated in degraded soil AGRIAMBI
Santos,Adriana A.; Agustini,José A.; Maltoni,Katia L.; Cassiolato,Ana M. R..
ABSTRACT The aim of this study was to evaluate, through mycorrhization (root colonization and number of spores of arbuscular mycorrhizal fungi - AMF), leaf acid phosphatase and soil chemical characteristics, the effects of the addition of residues (macrophytes and ash), hydrogel and the reintroduction of microorganisms in a degraded area cultivated with jatropha. Degradation occurred when the surface soil was removed during the construction of a hydroelectric power plant. The experiment was set in a randomized block design, using a 2 x 2 x 4 factorial scheme, i.e., two inoculation treatments (with and without soil-inoculum), two hydrogel treatments (with and without) and four with the addition of residues (macrophytes - MAC, ash, MAC + ash and control,...

Addition of waste and introduction of microorganisms after 45 years of soil degradation Rev. Ciênc. Agron.
Santos,Adriana Avelino; Agustini,José Antônio; Maltoni,Kátia Luciene; Cassiolato,Ana Maria Rodrigues.
ABSTRACT The construction of hydroelectric power plants (HPP) may result in environmental problems, such as extensive areas of exposed subsoil and conditions of extreme degradation. These areas require alternative that minimize impact and allow partial recovery of their ecosystem functions and vegetation. This study aimed to evaluate the effects of residue addition (organic/macrophytes - OR and inorganic/ash - AR), hydrogel, and inoculation of microorganisms in degraded soil, cultivated with Jatropha curcas, through fertility and microbial activity. A conserved Cerrado ("savannah") soil was the source of microorganisms - mainly mycorrhizal fungi. The experiment was conducted for 12 months (during 2010/2011) at the farm of UNESP-School of Engineering/Campus...

An in vitro study of two GAG-like marine polysaccharides incorporated into injectable hydrogels for bone and cartilage tissue engineering ArchiMer
Rederstorff, Emilie; Weiss, Pierre; Sourice, S.; Pilet, P.; Xie, F.; Sinquin, Corinne; Colliec-jouault, Sylvia; Guicheux, Jerome; Laib, S..
Natural polysaccharides are attractive compounds with which to build scaffolds for bone and cartilage tissue engineering. Here we tested two non-standard ones, HE800 and GY785, for the two-dimensional (2-D) and three-dimensional (3-D) culture of osteoblasts (MC3T3-E1) and chondrocytes (C28/I2). These two glycosaminoglycan-like marine exopolysaccharides were incorporated into an injectable silylated hydroxypropylmethylcellulose-based hydrogel (Si-HPMC) that has already shown its suitability for bone and cartilage tissue engineering. Results showed that, similarly to hyaluronic acid (HA) (the control), HE800 and GY785 significantly improved the mechanical properties of the Si-HPMC hydrogel and induced the attachment of MC3T3-E1 and C28/I2 cells when these...

Biomechanical and histological evaluation of hydrogel implants in articular cartilage BJMBR
Malmonge,S.M.; Zavaglia,C.A.C.; Belangero,W.D..
We evaluated the mechanical behavior of the repaired surfaces of defective articular cartilage in the intercondylar region of the rat femur after a hydrogel graft implant. The results were compared to those for the adjacent normal articular cartilage and for control surfaces where the defects remained empty. Hydrogel synthesized by blending poly(2-hydroxyethyl methacrylate) and poly(methyl methacrylate-co-acrylic acid) was implanted in male Wistar rats. The animals were divided into five groups with postoperative follow-up periods of 3, 5, 8, 12 and 16 weeks. Indentation tests were performed on the neoformed surfaces in the knee joint (with or without a hydrogel implant) and on adjacent articular cartilage in order to assess the mechanical properties of...

Effect of drying method on mechanical, thermal and water absorption properties of enzymatically crosslinked gelatin hydrogels Anais da ABC (AABC)
SIMONI,RAYSSA C.; LEMES,GISLAINE F.; FIALHO,SANDRA; GONÇALVES,ODINEI H.; GOZZO,ANGELA M.; CHIARADIA,VIVIANE; SAYER,CLAUDIA; SHIRAI,MARIANNE A.; LEIMANN,FERNANDA V..
ABSTRACT Enzymatically crossliked gelatin hydrogel was submitted to two different drying methods: air drying and freeze drying. The resulting polymeric tridimensional arrangement (compact or porous, respectively) led to different thermal and swelling properties. Significant differences (p < 0.05) on thermal and mechanical characteristics as well as swelling in non-enzymatic gastric and intestinal simulated fluids (37 ºC) were detected. Water absorption data in such media was modelled according to Higuchi, Korsmeyer-Peppas, and Peppas-Sahlin equations. Freeze dried hydrogel showed Fickian diffusion behavior while air dried hydrogels presented poor adjustment to Higuchi model suggesting the importance of the relaxation mechanism at the beginning of...

Effects of nanoparticles of hydroxy-aluminum phthalocyanine on markers of liver injury and glucose metabolism in diabetic mice BJMBR
Melo,M.A.B.; Caetano,W.; Oliveira,E.L.; Barbosa,P.M.; Rando,A.L.B.; Pedrosa,M.M.D.; Godoi,V.A.F..
Photodynamic therapy, by reducing pain and inflammation and promoting the proliferation of healthy cells, can be used to treat recurrent lesions, such as diabetic foot ulcers. Studies using the photosensitizer phthalocyanine, together with the nanostructured copolymeric matrix of Pluronic® and Carbopol® for the treatment of diabetic foot ulcers and leishmaniosis lesions, are showing promising outcomes. Despite their topical or subcutaneous administration, these molecules are absorbed and their systemic effects are unknown. Therefore, we investigated the effect of the subcutaneous administration of the hydroxy-aluminum phthalocyanine hydrogel without illumination on systemic parameters, markers of liver injury, and liver energy metabolism in type 1 diabetic...

Enriching a cellulose hydrogel with a biologically active marine exopolysaccharide for cell-based cartilage engineering ArchiMer
Rederstorff, E.; Rethore, G.; Weiss, P.; Sourice, S.; Beck-cormier, S.; Mathieu, E.; Maillasson, M.; Jacques, Y.; Colliec-jouault, Sylvia; Fellah, B. H.; Guicheux, J.; Vinatier, C..
The development of biologically and mechanically competent hydrogels is a prerequisite in cartilage engineering. We recently demonstrated that a marine exopolysaccharide, GY785, stimulates the in vitro chondrogenesis of adipose stromal cells. In the present study, we thus hypothesized that enriching our silated hydroxypropyl methylcellulose hydrogel (Si-HPMC) with GY785 might offer new prospects in the development of scaffolds for cartilage regeneration. The interaction properties of GY785 with growth factors was tested by surface plasmon resonance (SPR). The biocompatibility of Si-HPMC/GY785 towards rabbit articular chondrocytes (RACs) and its ability to maintain and recover a chondrocytic phenotype were then evaluated in vitro by MTS assay, cell counting...

Manipulation of a quasi-natural cell block for high-efficiency transplantation of adherent somatic cells BJMBR
Chung,H.J.; Hassan,M.M.; Park,J.O.; Kim,H.J.; Hong,S.T..
Recent advances have raised hope that transplantation of adherent somatic cells could provide dramatic new therapies for various diseases. However, current methods for transplanting adherent somatic cells are not efficient enough for therapeutic applications. Here, we report the development of a novel method to generate quasi-natural cell blocks for high-efficiency transplantation of adherent somatic cells. The blocks were created by providing a unique environment in which cultured cells generated their own extracellular matrix. Initially, stromal cells isolated from mice were expanded in vitro in liquid cell culture medium followed by transferring the cells into a hydrogel shell. After incubation for 1 day with mechanical agitation, the encapsulated cell...

Modeling drying of seed maize using super absorbent hydrogel under hermetic conditions CIGR Journal
Kiburi, Florence Gatwiri; Kanali, |Christopher; Mbuge, Duncan Onyango.
The objective of this study was to establish the feasibility of applying the Page, Two term exponential, Newton, Logarithmic and Henderson and Pabis mathematical drying models for predicting the drying kinetics of seed maize using super absorbent hydrogel under hermetic conditions. To achieve this, the study was conducted using hydrogel to seed maize ratios by weight of 0:1, 1:5, 1:10 and 1:15 at four different drying temperature levels of 25oC, 30oC, 35oC and 40oC at different initial seed maize moisture contents of 16%, 28% and 53% (dry basis). The moisture data obtained were fitted into the five drying models using non-linear regression analysis (MS Excel 2003TM) based on the minimization of sum of squares by adjusting the model constants. The...

WATER-RETAINING POLYMER MITIGATES THE WATER DEFICIT IN Schinus terebinthifolia: PHOTOSYNTHETIC METABOLISM AND INITIAL GROWTH REA
Beltramin,Francisco A.; Silva,Willian C.; Santos,Cleberton C.; Scalon,Silvana de P. Q.; Vieira,Maria do C..
ABSTRACT The use of water-retaining polymer (hydrogel) can mitigate the damage to the photosynthetic apparatus and increase the robustness of young plants under water deficit conditions. Thus, the aim of this study was to evaluate the effect of water-retaining polymer on the morphophysiological aspects of Schinus terebinthifolia Raddi submitted to water deficit. The studied water regimes were characterized by three plant irrigation management: 1) control: irrigated daily, 2) stress: water deficit (irrigation suspension) and 3) stress + hydrogel: water deficit + water-retaining polymer. The plants were submitted to irrigation suspension until photosynthesis approached zero [1st P0], followed by resumption of irrigation (REI) for 15 days. After REI, until...
